Abstract
Introduction. Overcoming resistance of malignant neoplasms to therapy is one of the most pressing problems of modern healthcare. Mitochondrial uncouplers have shown efficacy in various diseases and are being actively studied for the purposes of antitumor therapy.
Aim. To study the effects of platinum (II) carbene complexes causing depolarization of inner mitochondrial membrane on glioblastoma cells with varying initial sensitivity to chemotherapy for evaluation of possibility of using mitochondrial uncouplers for overcoming drug resistance in this tumor.
Materials and methods. Cytotoxic effect was evaluated using MTT assay, mitochondrial potential level using JC-1 cation dye, proliferative activity of the cells using 5-ethynyl-2’-deoxyuridine with iF555 fluorescent probe. Statistical data processing was performed in RStudio using R programming language.
Results. Comparable cytotoxic effect of platinum (II) carbene complexes in А172 and T98G glioblastoma cell lines was observed. Treatment of both lines causes statistically significant changes in the condition of inner mitochondrial membrane. Moreover, one of the compounds was more effective in the T98G cell line which is considered to be more resistant to chemical agents. All mitochondrial uncouplers caused statistically significant decrease in proliferative activity.
Conclusion. Platinum (II) carbene complexes have cytotoxic effect in glioblastoma cells with varying initial sensitivity to chemotherapy. Additionally, high efficacy and promise of these compounds as potential agents for overcoming resistance were demonstrated.
