Our previous studies for some ABC genes<italic> </italic>have established that the expression of these genes correlates with the effectiveness of neoadjuvant chemotherapy (NAC). Some of the clinical<italic> </italic>studies indicate that ABC transporters can influence not only the formation of chemoresistance in the tumor, but also the progression, invasion and metastasis of the tumor node. <italic> </italic><bold>Objective:</bold> to evaluate the level of transcription of all 49 known ABC genes in a breast tumor before and after treatment and their prognostic<italic> </italic>significance.<italic> </italic><bold>Materials and methods.</bold> The study included 31 patients with a diagnosis of stage IIA – IIIB breast cancer. RNA was isolated from paired<italic> </italic>samples of tumor tissue before and after NAC. A microarray study of all tumor samples was performed on Clariom<sup>ТМ</sup> S Assay, human microarrays. Using microarray studies, the expression of 49 genes of the ABC transporter family was studied. Analysis of the microchip data was<italic> </italic>carried out using the program Transcriptome Analysis Console (TAC) software 4.0.<italic> </italic><bold>Results.</bold> It was found that changes in the expression (increase/decrease during NAC) of the ABCA5, ABCA7, ABCB1, ABCB4, ABCB11,<italic> </italic>ABCC1, ABCC10, ABCC11, ABCG1, ABCG2, ABCG4, ABCG5, ABCG8 genes are statistically significantly associated with the response to<italic> </italic>NAC. In addition, the prognostic significance of ABCB1 and ABCB4 gene expression was established. Survival analysis showed that 5-year<italic> </italic>survival rates in patients with high gene expression of ABCB1 and ABCB4 are lower compared to patients with low expression of these genes<italic> </italic>(log-rank-test p = 0.001 and 0.04 respectively).<italic> </italic><bold>Conclusion.</bold> Data were obtained on the relationship of gene expression of the ABC transporter family with the effect of NAC in patients with<italic> </italic>breast cancer and the outcome of the disease. The prognostic potential of the ABCB1 and ABCB4 genes in patients with breast cancer has<italic> </italic>been established.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Одной из основных причин неэффективности химиотерапии по-прежнему считают формирование фенотипа множественной лекарственной устойчивости опухоли за счет экспрессии энергозависимых белков АВС-транспортеров. Результаты<italic> </italic>предыдущих наших исследований для некоторых генов ABC позволили установить, что экспрессия данных генов коррелирует с эффективностью неоадъювантной химиотерапии (НХТ). Стоит отметить, что некоторые из клинических исследований указывают на то, что ABC-транспортеры могут влиять не только на формирование химиорезистентности в опухоли, но также<italic> </italic>на прогрессию, инвазию и метастазирование опухолевого узла.<italic> </italic><bold>Цель исследования</bold> – оценка уровня транскриптов всех известных 49 генов ABC в опухоли молочной железы до и после проведения<italic> </italic>лечения и их прогностической значимости.<italic> </italic><bold>Материалы и методы.</bold> В исследование была включена 31 пациентка с диагнозом рака молочной железы IIA–IIIB стадий. РНК<italic> </italic>выделяли из парных образцов опухолевой ткани до и после НХТ. Проведено микроматричное исследование всех образцов опухоли<italic> </italic>на микрочипах Clariom<sup>ТМ</sup> S Assay, human. С помощью микроматричного исследования изучена экспрессия 49 генов семейства<italic> </italic>ABC-транспортеров. Анализ данных микрочипа осуществляли с использованием программы Transcriptome Analysis Console (TAC)<italic> </italic>software 4.0.<italic> </italic><bold>Результаты.</bold> Установлено, что изменение экспрессии (повышение/снижение в процессе НХТ) генов ABCA5, ABCA7, ABCB1, ABCB4,<italic> </italic>ABCB11, ABCC1, ABCC10, ABCC11, ABCG1, ABCG2, ABCG4, ABCG5, ABCG8 статистически значимо связано с ответом на НХТ.<italic> </italic>Кроме этого, установлена прогностическая значимость экспрессии генов ABCB1 и ABCB4. Анализ выживаемости продемонстрировал, что показатели 5-летней выживаемости у больных с высокой экспрессией генов ABCB1 и ABCB4 ниже по сравнению таковыми у пациенток с низкой экспрессией данных генов (log-rank-тест p = 0,001 и 0,04 соответственно).<italic> </italic><bold>Заключение.</bold> Получены данные о связи экспрессии генов семейства ABC-транспортеров с эффективностью НХТ у больных раком<italic> </italic>молочной железы и исходом заболевания.
