Tumor progression is also caused by the aberrant activity of intercellular signaling pathways, which can arise due to mutations in genes encoding their components. In particular, the oncogenic role of NOTCH1 receptor of Notch signaling pathway has been proven for various cancer types, including lung and colorectal cancers. In this research, we delved deeper into the importance of NOTCH1 receptor expression for the progression of these malignancies.</p><p><bold>   Aim. </bold>To investigate the importance of NOTCH1 expression in maintaining the cancer stem cell (CSC) pool and phenotype of human lung and colon cancers.</p><p><bold>   Materials and methods. </bold>Experiments were performed on previously obtained NOTCH1 knockdown cell lines of human lung A549 and colon HCT116 carcinomas. First of all, we studied the effect of NOTCH1 knockdown on the metastatic ability of A549 cells and the tumorigenicity of A549 and HCT116 cells when injected to immunodeficient BALB/c nu/nu mice. Next, we carried out in vitro tests to determine CSC pool and phenotype in tumor cell culture: cytofluorimetric analysis of ABC-transporters activity to exclude dye to the external environment and analysis of colony formation in a semi-liquid medium. In conclusion, we assessed the proportion of cells in the culture producing the CSC marker – CD133 using flow cytometry and the expression level of some genes associated with CSC phenotype (NANOG, POU5F1, SOX2) using real-time polymerase chain reaction.</p><p><bold>   Results. </bold>NOTCH1 knockdown decreased the number of experimental animals with metastases, the number of formed metastatic foci and increased in the minimum cell inoculation dose. The activity of ABC-transporters, the ability for unattached growth, the proportion of CD133-positive cells in culture, and the expression of genes associated with maintaining of CSC pool and phenotype decreased under NOTCH1 knockdown in both cell lines.</p><p><bold>   Conclusion. </bold>NOTCH1 expression is important for maintaining CSC pool and phenotype of human lung and colon carcinomas. The obtained data may be valuable in the development of anticancer therapies.</p></abstract><trans-abstract xml:lang="ru"><p><bold>   Введение.</bold> Рак легкого и колоректальный рак являются одними из самых распространенных типов злокачественных новообразований, характеризующихся неблагоприятным прогнозом. Опухолевая прогрессия обусловливается аберрантной активностью межклеточных сигнальных путей, которая может возникать за счет мутаций в генах, кодирующих их компоненты. В частности, онкогенная роль рецептора NOTCH1 сигнального пути Notch доказана для различных типов рака, включая рак легкого и колоректальный рак. В рамках настоящего исследования мы изучали важность продукции рецептора NOTCH1 для прогрессии данных типов злокачественных новообразований.</p><p><bold>   Цель исследования</bold> – изучение роли экспрессии NOTCH1 в поддержании пула и фенотипа опухолевых стволовых клеток (ОСК) карцином легкого и толстой кишки человека.</p><p><bold>   Материалы и методы.</bold> Эксперименты проводили на полученных ранее клеточных линиях карцином легкого А549 и толстой кишки HCT116 человека с подавленной экспрессией NOTCH1. В первую очередь было изучено влияние подавления экспрессии NOTCH1 на способность к метастазированию клеток А549 и туморогенность клеток А549 и НСТ116 при их введении иммунодефицитным бестимусным мышам линии BALB/c nu/nu. Далее были проведены тесты in vitro по определению пула и фенотипа ОСк в культуре опухолевых клеток: цитофлуориметрический анализ активности АВС-транспортеров (выявление способности к выведению красителя во внешнюю среду) и анализ колониеобразования в полужидкой среде. В заключение определены доля клеток в культуре, продуцирующих маркер ОСК (CD133), методом проточной цитофлуориметрии и уровень экспрессии некоторых генов, продукты которых ассоциированы с фенотипом ОСк (NANOG, POU5F1, SOX2), с помощью полимеразной цепной реакции в реальном времени.</p><p><bold>   Результаты.</bold> Подавление экспрессии NOTCH1 приводило к снижению числа экспериментальных животных с метастазами и количества сформированных метастатических очагов, а также к увеличению минимальной прививочной дозы. Активность АВС-транспортеров, способность к неприкрепленному росту, доля CD133-положительных клеток в культуре и экспрессия генов, продукты которых связаны с поддержанием пула и фенотипа ОСк, снижались при подавлении экспрессии NOTCH1 в обеих клеточных линиях.</p><p><bold>   Заключение. </bold>Экспрессия гена, кодирующего рецептор NOTCH1, важна для поддержания пула и фенотипа ОСК карцином легкого и толстой кишки человека.
