In glial tumors, lipid metabolism becomes abnormal. Analysis of lipid metabolism components can be an important characteristic of molecular and genetic profile of gliomas.<bold>Aim</bold>. To determine the correlation between plasma lipidome profile and immunohistochemical characteristics of glial tumors and to evaluate clinical significance of blood lipid spectrum analysis in preoperative assessment of molecular profile of gliomas.<bold>Materials and methods</bold>. Immunohistochemical measurement of O-6-methylguanine-DNA-methyl transferase (MGMT), Ki-67, p53, IDH1 tumor markers was performed using the corresponding antibody clones. Composition of plasma lipids was assessed using thin layer chromatography.<bold>Results</bold>. Even at the early stages of gliomagenesis, significant differences in cholesterol ethers, lysophosphatidylcholines, phosphatidylcholine (PC)/ lysophosphatidylcholine (LPC) ratio, neutral lipids (NL)/phospholipids (PL) in the blood were observed. Significant correlations between Ki-67, MGMT tumor markers and the above-mentioned lipidome parameters were found. The PC/LPC, NL/PL ratios in the blood of the patients from the groups with higher (above 10 %) and lower (below 10 %) Ki-67 mitotic indexes compared to healthy individuals were significantly lower. Therefore, the values of lipidome parameters allow to indirectly assess proliferative activity of gliomas which can be used for preoperative diagnosis of these tumors. No significant differences in the plasma PC/LPC and NL/PL ratios were found between the groups with MGMT promoter methylation and without it. No indirect predictor criteria for MGMT were found.<bold>Conclusion</bold>. It is impossible to determine decreased epigenetic activity of corresponding transcripts and preoperative prognosis for alkylating agent therapy based on the parameters of plasma lipid metabolism.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение</bold>. При глиальных опухолях обмен липидов нарушается. Анализ компонентов липидного обмена может быть важной характеристикой молекулярно-генетического профиля глиом.<bold>Цель исследования</bold> – определить взаимосвязь параметров липидома плазмы и иммуногистохимических характеристик глиальных опухолей и оценить клиническую значимость анализа липидного спектра крови для дооперационной оценки молекулярного профиля глиом.<bold>Материалы и методы</bold>. Иммуногистохимическое определение уровня онкомаркеров О-6-метилгуанин-ДНК-метилтрансферазы (MGMT), Ki-67, p53, IDH1 проводили с использованием клонов соответствующих антител. Состав липидов плазмы крови анализировали методом тонкослойной хроматографии.<bold>Результаты</bold>. Уже на начальных стадиях глиомагенеза выявлены значимые различия уровней эфиров холестерола, лизофосфатидилхолинов, соотношений фосфатидилхолин (ФХ)/лизофосфатидилхолин (ЛФХ), нейтральные липиды (НЛ)/фосфолипиды (ФЛ) в плазме крови. Показаны значимые корреляционные зависимости онкомаркеров Ki-67, MGMT от вышеназванных параметров липидома. Соотношения ФХ/ЛФХ, НЛ/ФЛ в плазме крови пациентов были значимо ниже в группах с высоким (более 10 %) и низким (менее 10 %) митотическими индексами Ki-67 по сравнению со здоровыми людьми. Таким образом, значения параметров липидома позволяют опосредованно судить о пролиферативной активности глиом, что может быть использовано для дооперационной диагностики этих образований. Значимых различий соотношений ФХ/ЛФХ и НЛ/ФЛ в плазме крови между группами с метилированием промотора MGMT и его отсутствием не обнаружено. Критериев-предикторов опосредованного выявления метилирования промотора MGMT не выявлено.<bold>Заключение</bold>.
