BC heterogeneity in large part is determined by unique epigenetic landscape created by various factors. However, despite obvious heterogeneity, modern therapeutic approaches in this disease are limited to evaluation of histological subtype, receptor status (estrogen, progesterone, human epidermal growth factor 2 receptors), and proliferative activity (Ki-67). All important oncogenes and tumor suppressor genes, as well as tumor microenvironment, are controlled by epigenetic factors, and study of these control mechanisms and identification of clinically significant epigenetic biomarkers is an important problem. Currently, there are thousands of studies on epigenetic regulation in BC, systematization of which will help to better understand complex mechanisms of carcinogenesis.</p> <p>This review describes the main epigenetic mechanisms including DNA and RNA methylation, histone post-translational modification, and regulatory activity of non-coding RNAs. Characteristics of epigenetic regulation of various molecular and biological BC subtypes taking into account diagnostic and prognostic markers are analyzed, potential therapeutic targets are evaluated. The article also considers signaling pathways regulated by epigenetic modulators.</p>
В статье также рассматриваются сигнальные пути, которые подвергаются регуляции со стороны эпигенетических модуляторов.</p></trans-abstract><kwd-group xml:lang="en"><kwd>breast cancer</kwd><kwd>epigenetics</kwd><kwd>oncogenesis</kwd><kwd>DNA methylation</kwd><kwd>histone modification</kwd><kwd>long noncoding RNA</kwd><kwd>microRNA</kwd><kwd>tumor microenvironment</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>рак молочной железы</kwd><kwd>эпигенетика</kwd><kwd>онкогенез</kwd><kwd>метилирование ДНК</kwd><kwd>модификации гистонов</kwd><kwd>длинная некодирующая РНК</kwd><kwd>микроРНК</kwd><kwd>опухолевое микроокружение</kwd></kwd-group><funding-group><award-group><funding-source><institution-wrap><institution xml:lang="ru">Министерство науки и высшего образования Российской Федерации</institution></institution-wrap><institution-wrap><institution xml:lang="en">Ministry of Science and Higher Education of the Russian Federation</institution></institution-wrap></funding-source><award-id>075-03-2026-217/2</award-id></award-group><funding-statement xml:lang="en">The study was carried out with the support of the Ministry of Education and Science of Russia has approved the project FZEG-2026-0008 “Research of the fundamental mechanisms of malignant neoplasms development to improve molecular and genetic methods of personalized diagnostics by identifying new targets for targeted therapy, predictors of resistance, and clinically significant molecular and biological markers in pathomorphology”.</funding-statement><funding-statement xml:lang="ru">Работа поддержана проектом Минобрнауки России FZEG-2026-0008 «Исследование фундаментальных механизмов развития злокачественных новообразований для совершенствования молекулярно-генетических методов персонализированной диагностики путем выявления новых точек приложения таргетной терапии, предикторов резистентности и клинически значимых молекулярно-биологических маркеров в патоморфологии».</funding-statement></funding-group></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Kang Z., Wang J., Liu J. et al.
