Loss of NSD1 is associated with improved prognosis in HPV (human papilloma virus) negative cancer. However, data on the signaling pathways regulated by NSD1 are limited.</p> <p><bold>Aim.</bold> Validation and systemic assessment of reproducibility of the NSD1-Akt/mTORC1 axis based on integrative analysis of open transcriptomic and proteomic data from cell lines and samples of head and neck squamous cell carcinoma.</p> <p><bold>Materials and methods.</bold> Systemic analysis was performed of the connection between NSD1 and Akt/mTORC1 signaling pathway using open transcriptomic and proteomic data (panel of squamous cell carcinoma cell lines on the DepMap portal and clinical cohorts from the The Cancer Genome Atlas (TCGA)). Pathway activity was evaluated using ssGSEA/GSEA (single-sample Gene Set Enrichment Analysis), proteomic activity – using protein expression data obtained with Reverse Phase Protein Array (RPPA), statistical significance – using standard correlation and nonparametric tests.</p> <p><bold>Results.</bold> In the cell line panel, elevated NSD1 expression correlated with stronger PI3K/AKT/mTOR transcription signature and increased phosphorylation of the key pathway components according to RPPA data. Similar correlation was observed in patient tumor samples. Data on isogenic cell lines with <italic>NSD1</italic> knockout demonstrated decreased expression of mTORC1 target genes.</p> <p><bold>Conclusion.</bold> Reproducible correlation between NSD1 level and Akt/mTORC1 signaling pathway activity in head and neck cancer was established. However, additional confirmation of the NSD1 role in regulation of the Akt/mTORC1 pathways requires <italic>in vitro</italic> and <italic>in vivo</italic> investigation of expression and phosphorylation of protein components of this cascade, as well as functional experiments to evaluate proliferation, survival and drug sensitivity of head and neck cancer cells. Clinical significance of the established correlation in the context of disease progression requires further verification.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Гистоновая метилтрансфераза NSD1 часто инактивирована при плоскоклеточном раке головы и шеи. Потеря NSD1 ассоциируется с улучшенным прогнозом при отрицательном ВПЧ-статусе (ВПЧ – вирус папилломы человека). Однако данные о сигнальных путях, регулируемых NSD1, ограниченны.</p> <p><bold>Цель исследования</bold> – валидация и системная оценка воспроизводимости сигнальной оси NSD1-Akt/mTORC1 на основе интегративного анализа общедоступных транскриптомных и протеомных данных относительно клеточных линий и образцов плоскоклеточного рака головы и шеи.</p> <p><bold>Материалы и методы.</bold> Проведен системный анализ связи NSD1 с сигнальным путем Akt/mTORC1 на основе доступных транскриптомных и протеомных данных (панель клеточных линий плоскоклеточного рака головы и шеи на портале DepMap и клинические когорты проекта «Атлас ракового генома» (The Cancer Genome Atlas, TCGA)). Активность путей оценивали с помощью ssGSEA/GSEA (single-sample Gene Set Enrichment Analysis), протеомную активность – по данным об экспрессии белков, которые были получены методом обратно-фазных белковых микрочипов (Reverse Phase Protein Array, RPPA), статистическую значимость – с использованием стандартных корреляционных и непараметрических тестов.</p> <p><bold>Результаты.</bold> В панели клеточных линий повышенная экспрессия NSD1 коррелировала с усилением транскрипционной сигнатуры PI3K/Akt/mTOR и увеличением фосфорилирования ключевых компонентов пути по данным RPPA. Аналогичная ассоциация наблюдалась в опухолевых образцах пациентов. Данные по изогенным клеточным линиям с нокаутом <italic>NSD1</italic> продемонстрировали снижение экспрессии генов-мишеней mTORC1.</p> <p><bold>Заключение.</bold> Выявлена воспроизводимая связь между уровнем NSD1 и активностью сигнального пути Akt/mTORC1 при раке головы и шеи. Однако для дополнительного подтверждения роли NSD1 в регуляции пути Akt/mTORC1 необходимы исследования экспрессии и фосфорилирования белковых компонентов данного каскада <italic>in vitro</italic> и <italic>in vivo</italic>, а также функциональные эксперименты по оценке пролиферации, выживаемости и лекарственной чувствительности клеток рака головы и шеи.
